为什么使用hilic?
尽管反相液相色谱（rplc）是至今应用zui广的色谱分离技术，它能与各种常规的检测方法结合，解决多种分析应用问题，但对某些分析物，特别是极性和亲水性化合物却无法或很少保留。*以来，人们采用正相液相色谱（nplc），并使用不利环保的非水溶性流动相如己脘或环己脘来分析这些物质。
但是在这种实验条件下，很多极性和亲水化合物往往很难溶解，从而限制了nplc的应用范围。从现在起有了另一种新的解决办法，即选用亲水作用液相色谱-hilic。
什么是亲水作用色谱（hilic）：
亲水作用液色谱（hydrophilic interaction liquidchromatography，hilic） 是一种直接分离极性和亲水性化合物的色谱技术，为了便于理解，我们可以把它看作为是一种反相色谱溶剂做为流动相的正相色谱。
因此hilic是：
●亲水性的固定相
●以水、有机溶剂和缓冲液作为流动相洗脱剂
常见的hilic方法采用50-90%有机溶剂和一定比例的水，并且含有低浓度的缓冲盐。如乙腈/水/醋酸铵体系
在hilic方法中，洗脱顺序和反相色谱正好相反，即极性越大，保留时间越长。
